YEAR: 1989 CITATION: J Agric Food Chem, 37(4), 998-1005 [English] FDA #: F17893 ABSTRACT: Bulk com. grain, such as soybeans and wheat, may be contaminated by nongrain impurities, including jimson weed seeds, that coexist with the crop to be harvested. The content of the major alkaloids of jimson weed seeds: atropine and scopolamine, as well as protein, carbohydrate, fat, mineral, hemagglutinin, and tannin were detd. Combined GC-MS anal. of a seed ext. revealed the presence of atropine and scopolamine plus possibly 3 addnl. tropane-like alkaloids. An improved HPLC procedure (C8 Ultrasphere column with H2O-MeOH (64:36) contg. 0.02M pH 3 phosphate buffer and 0.01M Bu2NH counterion, detection at 200 nm) showed that the alkaloid concn. in samples obtained from different parts of the United States varied by as much as 50%: 1.69-2.71 mg/g for atropine and 0.36-0.69 mg/g for scopolamine. A strongly fluorescent green compd. of unknown structure also was present. Baking expts. with jimson weed seed-fortified wheat flour showed that atropine and scopolamine largely survive bread-baking conditions. Jimson weed seeds do not contain protease or amylase inhibitors. The relation of seed compn. to biol. effects in animals and the possible significance of low levels of the seeds in feed of food-producing animals and in the human diet are discussed
